Endeavor’s Phase IIb clinical for IPF met endpoint and the Hedgehog signaling pathway as a targe...

Endeavor BioMedicines recently announced that its IPF candidate taladegib (ENV-101) met the primary endpoint and multiple key secondary endpoints in a Phase IIb study. These results highlight the potential of taladegib in the treatment of idiopathic pulmonary fibrosis (IPF) and further validate the strategy of targeting the Hedgehog (Hh) signaling pathway. Current standard-of-care therapies, nintedanib and pirfenidone, can only slow the decline in lung function and do not halt or reverse disease progression. Taladegib, by inhibiting aberrantly activated Hedgehog signaling, reduces pulmonary fibrosis and improves lung function.

As a Hedgehog pathway inhibitor with the same target, GT1708F also has therapeutic potential for IPF. It inhibits SMO protein activity, thereby affecting Hh pathway activity and the expression of downstream related proteins. Hh pathway reactivation is a characteristic of fibrotic lung tissue in IPF and influences fibroblast migration and proliferation. Notably, Hedgehog inhibitors not only hold promise for the treatment of IPF but also have development value in other fibrotic diseases. In in vitro studies, GT1708F demonstrated superior inhibitory activity against the Hh pathway compared with taladegib. In in vivo IPF models, GT1708F effectively ameliorated pulmonary fibrosis and was superior to taladegib at the same dose. Currently, GT1708F has completed a phase I clinical trial in China for hematologic malignancies and has been authorized to initiate  a phase II clinical trial in China and a phase I clinical trial in the US for IPF.

As a great health company in innovative topical dermatological drugs, we are open to global collaborative development of GT1708F and look forward to in-depth discussions with more partners on collaboration models such as rights licensing, including but not limited to license-out and the formation of a NewCo. We hope to work together with partners to accelerate clinical development and bring innovative therapies to patients worldwide as soon as possible.
